Use it for everyday accidents like stepping on a toe or dropping a pen. It is perfect for interrupting someone politely to ask a question. Use it when you realize you've made a mistake in a conversation. It is also the standard way to apologize to friends, family, and colleagues. Basically, if you feel a tiny bit of guilt, say it.

Don't use desculpa for very serious life events, like a deep betrayal or a major tragedy. In those cases, sinto muito (I am very sorry/I feel much) is more appropriate. Also, avoid using the informal desculpa with your boss or a judge. For high-formality situations, you need to switch the ending to desculpe. Using the wrong one won't start a war, but it might feel a bit too 'buddy-buddy'.
